A. Antimetabolities
MTX : structural analog of folic acid
act as irreversible inhibitor of the active site of the
enz. dihydrofolate reductase(necessary for DNA synthesis)
5-FU : irreversible inhibitor of the enz.thymidylate synthetase
(necessary for the synthesis of thymidine)
pyrimidine 유사체
6-mercaptopurine : Hypoxanthine ribonucleotide (inosinic acid)를
adenine과 xanthine ribonucleotide로 전환시키는 효소
와 상경적으로 작용.
cytarabine : cytosine arabinoside라고도 하며, cell cycle의 G 1-phase
에서 S-phase에로의 과정에 선택적으로 작용하고,
DNA polymerase를 억제함으로써 DNA 합성을 저지.
enocitabine : cytarabine의 pro-drug이다.
tegafur : futrafur라고도 하며, 체내에서 fluorouracil을 방출시켜
작용발현.
carmofur : fluorouracil의 유도체로 유사한 작용발현.
